Overall objective of the project

As an outcome of the Euphresco project 2018-A-271, entitled ‘Rapid identification of plant-health related bacteria by MALDI-TOF mass spectrometry (MALD-ID)’ gaps in the availability of reference databases for plant pathogenic bacteria were partially filled. The project has resulted in the public release of 117 in-house created reference spectra that are compatible with the MALDI-TOF MS system as offered by Bruker. Several different species of the genera Clavibacter, Curtobacterium, Erwinia, Pantoea, Pectobacterium, Pseudomonas, Ralstonia and Xylophilus are represented in this dataset. The spectra were tested on the technical exchange possibilities but their well-functioning on third-party MALDI-TOF MS machines was poorly evaluated. The aim of this project is to evaluate in-house created Bruker-based databases, in particular for Clavibacter sepedonicus, Curtobacterium flaccumfaciens pv. flaccumfaciens, Pantoea stewartii subsp. stewartii, Ralstonia solanacearum, Ralstonia pseudosolanacearum, Pectobacterium sp. and Dickeya sp. on their usability for identification at third party laboratories.
